Antinuclear antibodies (ANA) are primarily used to detect autoimmune diseases, especially connective tissue diseases, such as systemic lupus erythematosus (SLE), Sjögren's syndrome (SS), polymyositis or dermatomyositis (PM/DM), and systemic sclerosis (SSc).

ANA refers to a group of autoantibodies produced against DNA, RNA, proteins, or molecular complexes of these substances within the cell nucleus. A positive result suggests the possible presence of an autoimmune disease. Different ANA components have distinct clinical implications—for example, anti-dsDNA antibodies indicate SLE activity, while anti-SS-A and anti-SS-B antibodies are helpful in diagnosing SS. Therefore, ANA testing is an important step in diagnosing autoimmune diseases.
