Oral administration is a systemic route of drug delivery. If adverse reactions occur, the medication should be discontinued promptly.
Potential adverse effects include the following:
1. Gastrointestinal reactions, such as nausea, vomiting, diarrhea, and abdominal pain;
2. Possible allergic reactions;
3. Hepatotoxicity, manifested by elevated hepatic transaminases, as well as dizziness and headache. Fluconazole is primarily excreted renally—approximately 80% is eliminated unchanged in the urine, with only a small fraction metabolized in the liver. Therefore, patients with impaired hepatic or renal function should avoid using fluconazole.
Its elimination half-life ranges from 27 to 37 hours, meaning its pharmacological effect persists for an extended duration. Consequently, dosing intervals can be relatively long. Specifically, for treating vulvovaginal candidiasis, the recommended regimen is 150 mg orally once every three days—i.e., twice weekly (e.g., Monday and Thursday). Each dose should be 150 mg.
