Motilium is domperidone tablets, an over-the-counter prokinetic agent primarily used to relieve symptoms of dyspepsia—including nausea, vomiting, belching, abdominal distension, and abdominal pain. Motilium should not be used for longer than one week, as prolonged use may lead to certain adverse effects. So, what are the actions and therapeutic effects of Motilium?
Actions and Therapeutic Effects of Motilium
Motilium—also known as domperidone tablets—is a prokinetic drug used to treat dyspepsia. “Prokinetic” refers to agents that enhance gastrointestinal motility. The primary mechanism of action of Motilium is as a peripheral dopamine receptor antagonist; it promotes gastrointestinal motility mainly by blocking dopamine receptors in the gastrointestinal tract.
However, if dopamine receptors in other parts of the body are also blocked, off-target (i.e., unintended) pharmacological effects—termed adverse reactions—may occur. The main adverse reactions associated with Motilium include the following:

1. Neurological adverse reactions: These commonly manifest as fatigue, dizziness, headache, or drowsiness following Motilium administration. Therefore, patients are advised to avoid activities requiring alertness—such as driving or operating machinery—while taking this medication.
2. Gastrointestinal adverse reactions: These may include abnormalities in liver function tests. Since domperidone is primarily metabolized in the liver, abnormal hepatic biochemistry suggests mild hepatic impairment, warranting cautious use. Domperidone is contraindicated in patients with moderate hepatic impairment.
3. Cardiac adverse reactions: Rarely, domperidone has been associated with cardiac arrhythmias and even sudden cardiac death. Thus, patients aged 60 years or older may face increased risk when using Motilium and should only take it under the supervision of a qualified physician. Additional adverse reactions may include dry mouth and skin rash.
Finally, we recommend adopting healthy dietary habits: minimize intake of raw, cold, greasy, or spicy foods; avoid overeating or binge eating; opt for smaller, more frequent meals; maintain regular physical activity; and ensure regular bowel movements. Regarding Motilium use, attention must be paid to both dosage and treatment duration: for acute vomiting, the recommended course is one week; for other indications, the maximum recommended duration is four weeks. If symptoms do not improve within these timeframes, prompt medical evaluation is advised.
