What Causes ALS?

Amyotrophic lateral sclerosis (ALS), commonly known as “frozen” or “locked-in” syndrome, is a rare neurological disorder characterized by progressive muscle atrophy and skeletal muscle weakness. In most cases, the exact cause remains unknown; however, potential contributing factors include genetic predisposition or exposure to environmental toxins.

What Causes ALS?

Amyotrophic lateral sclerosis (ALS) is a rare neurodegenerative disease. Its hallmark clinical features include skeletal muscle weakness, atrophy, and bulbar palsy. Immune-related factors play a role: clinical examinations frequently reveal abnormal immune function in ALS patients, including elevated levels of immune complexes and immunoglobulins. Deficiencies in trace elements—such as certain metals—are also commonly observed in individuals with ALS.

Genetic factors: Clinical studies indicate that ALS can be an autosomal inherited disorder. Depending on whether the causative gene is dominant or recessive, there is approximately a 5–10% chance of transmission to offspring. While the etiology of ALS is multifactorial, genetic abnormalities account for roughly 20% of cases. Environmental factors—including aluminum toxicity—also contribute to disease development in some patients.

For suspected ALS, patients should consult a neurologist for evaluation. Clinical assessment includes examination for signs of muscle atrophy; electromyography (EMG) may be performed when necessary to confirm diagnosis prior to initiating targeted therapy.
