The causes of lymph node nodule formation are varied, with common causes including:

1. Inflammatory infections: Bacterial, viral, fungal, and parasitic infections can all lead to lymphadenitis. For example, acute tonsillitis caused by streptococcus and reactive lymph node hyperplasia following viral infections; multiple inflammatory lesions in other areas such as oral inflammation, tonsillitis, and gingivitis.

2. Tumors: Malignant tumors, including lymphoma, leukemia, and metastatic cancers, can spread to lymph nodes through the lymphatic system, leading to the formation of lymph node nodules.

3. Metabolic diseases: Conditions such as hyperthyroidism and amyloidosis can also affect normal lymphatic system function, causing lymph node enlargement.

4. Drug reactions: Certain medications, such as penicillins, sulfonamides, and antiepileptic drugs, may cause drug-induced lymphadenopathy.

5. Cellular proliferative and metabolic disorders: Conditions such as lipid storage diseases and sarcoidosis may also cause lymph node enlargement, leading to the formation of lymph node nodules.
